   <subfield code="a">Đề tài đã đề xuất một phương pháp đơn giản và an toàn để tổng hợp belinostat và các dẫn xuất tương tự belinostat. Quy trình bắt đầu từ 3-nitrobenzaldehyde và bao gồm 5 bước phản ứng liên tiếp: (i) phản ứng ngưng tụ Knoevenagel được áp dụng để tổng hợp hợp chất 2a thông qua malonic acid (được xúc tác bởi pyridine) hoặc sự ngưng tụ của 3-nitrobenzaldehyde và anhydric acetic acid trong sự hiện diện của CH₃COONa để thu được hợp chất 2a1, (ii) phản ứng ester hóa, (iii) phản ứng khử nhóm nitro vòng thơm thành nhóm amino, (iv) sự giới thiệu nhóm chlorosulfonyl vào vòng thơm thông qua sự tạo thành muối diazonium và sự sulfon hóa sử dụng p-TsOH/NaNO₂ trong nước và thionyl chloride/CuCl trong nước (SOCl₂ trong H₂O để cung cấp SO₂); sự thế thân hạch với các amine, (v) phản ứng tạo hydroxamic acid bởi hydroxylamine. Belinostat (6a) được tổng hợp thành công với hiệu suất từ 12.0 - 14.0 % và hiệu suất của bảy dẫn xuất tương tự belinostat (6b-h) dao động từ 8.0 - 13.0 %. Trong đó, năm dẫn xuất được ký hiệu 6c, 6d, 6e, 6g và 6h là các dẫn xuất mới. Bảy dẫn xuất (6b-h) được sàng lọc khả năng gây độc trên các dòng tế bào ung thư (KB, Hep-G2, Lu-1 và MCF-7). Kết quả cho thấy chỉ có hợp chất 6b ức chế mạnh dòng tế bào ung thư gan (Hep-G2) và dòng tế bào ung thư phổi (Lu-1) với giá trị IC50 là 1.65±0.08 µg/mL và 0.59±0.04 µg/mL, so sánh với giá trị IC₅₀ của chất đối chứng dương Ellipticine lần lượt là 0.56±0.04 µg/mL và 0.40±0.05 µg/mL. Kết quả docking phân tử cho thấy các tương tác đáng chú ý giữa phối tử 6b và vị trí hoạt động của enzyme HDAC8 bao gồm liên kết cộng hóa trị với ion Zn²⁺nằm ở đáy của túi liên kết và các tương tác với các amino acid đặc trưng như Tyr306, His180, Gly151, Phe208, Phe152, Lys33. Năng lượng liên kết dự đoán là -6.62 kcal/mol.</subfield>
